Systems Biology Analyses Show Hyperactivation of Transforming Growth Factor-β and JNK Signaling Pathways in Esophageal Cancer.

Esophageal adenocarcinoma (EAC) is resistant to standard chemoradiation treatment, and some targeted therapies available. We use large-scale network profiles and pharmacogenetic analysis to identify signaling pathways regulated within the EAC network which may be targeted to slow tumor growth or progression.We 397 biopsy specimens collected from patients with EAC and nonmalignant Barrett’s esophagus (BE), with or without dysplasia.

We did the analysis of RNA-sequencing and used systems biology approaches to identify different pathways activated in EAC vs nonmalignant dysplastic tissue; Track activity was confirmed by immunohistochemistry and real-time quantitative polymerase chain reaction analysis of signal components in patient tissue samples. EAC humans (FLO-1 and EsoAd1), dysplastic BE (CP-B, CP-C, CP-D), and BE nondysplastic (CP-A) cells were incubated with pharmacological inhibitors or transfected with small interfering RNA.

We measure the effects on proliferation, colony formation, migration, and / or growth of tumor xenografts in nude EAC vs mice.Comparisons BE nondysplastic network show hyperactivation of transforming growth factor-β (TGFb) and / or June N-terminal kinase (JNK) pathway signal at more than 80% of the sample EAC. Immunohistochemical analysis showed increased nuclear localization of phosphorylated June and Smad proteins in tumor tissue EAC compared with nonmalignant tissue. Gene is regulated by the JNK pathway which TGFb and specifically expressed in EAC and dysplastic BE.

Pharmacological inhibition or knockdown of TGFb or JNK signaling components in EAC cells (FLO-1 or EsoAd1) significantly reduced cell proliferation, colony formation, cell migration, and / or growth of tumor xenografts in mice with SMAD4-independent manner. TGFb pathway inhibition in the cell lines BE reducing the proliferation of dysplastic, but not nondysplastic, cells.

In transcriptome analysis of the EAC and nondysplastic BE tissue, we found TGFb and JNK signaling pathways for hyperactivated in EACS and the genes regulated by this pathway will be expressed in the EAC and BE dysplastic. Inhibiting this pathway in EAC cells reducing their proliferation, migration, and the formation of xenograft tumors. Strategies for blocking TGFb signaling and JNK pathways may be developed for the treatment of EAC.
